Summary

The Leishman-Giemsa (LG) cocktail stain shows diagnostic efficiency comparable to the Papanicolaou (PAP) stain for oral squamous cell carcinoma (OSCC) screening. This cost-effective LG stain is a promising, less time-consuming alternative for exfoliative cytology.

Background:

  • Exfoliative cytology is a noninvasive method for detecting premalignant and malignant lesions.
  • Classical Papanicolaou (PAP) stain is standard, while Romanowsky stains like Leishman-Giemsa (LG) cocktail are underutilized in exfoliative cytology.
  • LG cocktail offers a potentially cost-effective, time-efficient staining technique with reduced infrastructural needs.

Purpose of the Study:

  • To compare the diagnostic efficiency of LG cocktail with PAP stain and Feulgen stain.
  • To evaluate cellular changes in mucosal cells indicative of oral squamous cell carcinoma (OSCC).
  • To assess the utility of LG stain in routine cytological procedures.

Main Methods:

  • Cytological smears were prepared from 10 healthy controls and 10 OSCC patients.
  • Smears were stained using LG cocktail, PAP stain, and Feulgen stain.
  • Staining characteristics and cytologic diagnoses were evaluated and statistically analyzed.

Main Results:

  • LG cocktail and PAP stain demonstrated nearly identical diagnostic results for OSCC.
  • Both LG cocktail and PAP stain were superior to Feulgen stain in diagnostic ability and staining characteristics.
  • LG cocktail exhibited favorable staining for nuclear and cytoplasmic details.

Conclusions:

  • The one-step LG cocktail stain is easy, cost-effective, and less time-consuming than PAP stain.
  • LG cocktail requires less infrastructural support compared to PAP stain.
  • Further evaluation of LG cocktail is recommended for oral cancer screening as a potential aid.
