Novel biochemical assays using RNase R, RNase H, and alkaline treatment combined with northern blotting are presented to study circular RNAs (circRNAs). These methods are particularly useful for analyzing circular RNA concatemers, which are difficult to study with traditional techniques.
Area of Science:
- Molecular Biology
- Genetics
- Biochemistry
Background:
- Circular RNAs (circRNAs) are a class of noncoding RNA found in eukaryotes.
- While most circRNAs are lowly expressed, some can be highly abundant.
- Current methods like RT-PCR are not always suitable for studying specific circRNA types.
Purpose of the Study:
- To establish and present biochemical assays for studying circRNAs.
- To specifically address the challenge of analyzing circular RNA concatemers.
- To provide reliable techniques for circRNA research.
Main Methods:
- Development of three biochemical assays: RNase R treatment, RNase H treatment, and alkaline treatment.
- Integration of these assays with northern blotting for detection and analysis.
- Application of these methods to study both general circRNAs and circular RNA concatemers.
Main Results:
- The presented biochemical assays are effective for studying circRNAs.
- These methods provide a means to analyze circular RNA concatemers, overcoming limitations of existing techniques.
- Northern blotting in conjunction with these treatments allows for robust circRNA characterization.
Conclusions:
- Biochemical assays combined with northern blotting offer a powerful approach for circRNA research.
- These methods are crucial for the detailed study of circular RNA concatemers.
- The established techniques advance the understanding and analysis of noncoding RNA species.

RNA Interference
This process occurs naturally in cells, often through the activity of genomically-encoded microRNAs. Researchers can take advantage of this mechanism by introducing synthetic RNAs to deactivate specific genes for research or therapeutic purposes. For example, RNAi could be used...
RNA Structure
The basic structure of RNA consists of a five-carbon sugar and one of four nitrogenous bases. Although most RNA is single-stranded, it can form complex secondary and tertiary structures. Such structures play essential roles in the regulation of transcription and translation.
Different Types of RNA Have the Same Basic Structure
There are three main types of ribonucleic acid (RNA): messenger RNA (mRNA), transfer RNA (tRNA), and ribosomal RNA (rRNA). All three RNA types consist of a...
