    Summary

    Sickle cell anemia (SS) erythrocytes show increased calcium (Ca2+) fluxes and a distinct, slowly exchangeable Ca2+ compartment compared to normal (AA) cells. This suggests excess Ca2+ in SS cells is compartmentalized and not readily available for cellular processes.

    Background:

    • Sickle cell anemia (SS) erythrocytes are known to have altered ion transport compared to normal (AA) erythrocytes.
    • Cytosolic ionized calcium ([Ca]i) levels and calcium (Ca2+) fluxes are critical parameters in red blood cell function and pathology.

    Purpose of the Study:

    • To investigate Ca2+ exchange fluxes and cytosolic ionized Ca2+ ([Ca]i) in SS and AA erythrocytes.
    • To determine the nature and accessibility of the exchangeable Ca2+ pool in SS cells.

    Main Methods:

    • Erythrocytes (AA and SS) were loaded with Ca2+ chelators (Quin2 or Benz2).
    • Ca2+ fluxes and [Ca]i were measured using 45Ca equilibration and release kinetics.
    • Chelator content was quantified using 45Ca binding and [3H]Quin2 incorporation.

    Main Results:

    • Chelator incorporation reduced ATP levels, with a smaller decrease in SS cells.
    • SS cells exhibited 30-75% higher Ca2+ fluxes than AA cells, but similar [Ca]i (~20 nM).
    • A second, slowly exchangeable Ca2+ compartment (3-5 µmol/L cells) was identified in SS cells, absent in AA cells.

    Conclusions:

    • The excess Ca2+ in SS cells is largely sequestered in a slowly exchangeable pool, limiting its availability for the Ca2+ pump and K+ channels.
    • This compartmentalization may be linked to intracellular vesicles or heterogeneity within the SS erythrocyte population.
