    Background:

    Purpose of the Study:

    • To investigate the impact of cell density on fragile X (fra(X)) expression.
    • To determine optimal culture conditions for maximizing fra(X) detection.
    • To improve diagnostic accuracy for fragile X syndrome.

    Main Methods:

    • Lymphoblast cell lines and whole blood from fra(X) individuals were cultured at varying densities (0.25 X 10(5) to 2 X 10(6) cells/ml).
    • Cultures were treated with FUdR (0.1 microM) 24 hours prior to harvest.

    • Optimizing culture conditions enhances the diagnostic yield for fragile X syndrome.
