Background:

  • The impact of enhancing SO during sleep on subsequent declarative memory encoding is not fully understood.

Purpose of the Study:

  • To investigate the effect of acoustically enhanced SO during a nap on the encoding of declarative memories.
  • To explore the relationship between SO enhancement, memory performance, and brain activity.

Main Methods:

  • Thirty-seven healthy young adults underwent two nap conditions: acoustic stimulation (STIM) and no stimulation (SHAM).
  • Auditory tones were phase-locked to SO up-states during naps in the STIM condition.
  • fMRI was used to measure brain activity during declarative memory encoding after naps, followed by recognition testing.

Main Results:

  • Acoustic stimulation successfully augmented SO, but no overall group benefit in memory performance was observed.
  • The degree of SO enhancement positively correlated with memory recollection and hippocampal activation during encoding.
  • Increased sleep spindle activity did not correlate with memory benefits or changes in hippocampal signals.

Conclusions:

  • Acoustic stimulation during naps can enhance declarative memory encoding, particularly when SO are augmented.
  • Hippocampal activation during encoding is positively associated with the degree of SO augmentation.
