    Background:

    • Progressive multiple sclerosis (MS) is associated with reduced activated suppressor cell function.
    • Previous studies indicated a defect in T8+ suppressor cells in MS patients.

    Purpose of the Study:

    • To investigate the persistence of T8+ suppressor cell dysfunction in multiple sclerosis.
    • To determine if the observed defect is intrinsic to T8+ cells or due to external factors.

    Main Methods:

    • Generated T8+ and T4+ cell lines from peripheral blood of MS patients and healthy controls.
    • Assessed suppressor cell activity using concanavalin A or OKT3 stimulation.
    • Measured [3H]thymidine uptake to evaluate cell proliferation.

    • T8+ cell-mediated suppressor activity was significantly reduced in MS patients (13%) compared to controls (68%).

    • The defect in T8+ activated suppressor cell function in MS is persistent.
