    Summary

    Lymph nodes near melanoma exhibit increased suppressor T-cell activity, impairing immune responses. This finding explains the reduced immune reactivity observed in regional lymph nodes close to the tumor.

    Background:

    • Lymph nodes near melanoma show heightened responses to immune stimuli.

    Purpose of the Study:

    • To investigate suppressor T-cell activity in lymph nodes at varying distances from primary melanoma.
    • To determine if suppressor T-cells contribute to diminished immune reactivity in regional lymph nodes.

    Main Methods:

    • Utilized a concanavalin A (Con A) suppressor cell assay on lymphocytes from tumor-free regional lymph nodes (proximal, intermediate, distal).
    • Assessed proliferation of autologous peripheral blood lymphocytes co-cultured with Con A-treated lymph node lymphocytes (LNL).
    • Measured T-lymphocyte subset ratios (T4:T8) and suppressor cell activity maintenance in culture with recombinant interleukin 2.

    Main Results:

    • Con A-treated LNL from nodes proximal to melanoma demonstrated greater suppression of peripheral blood lymphocyte response compared to distal nodes.
    • No significant changes in T4:T8 ratios were observed after Con A treatment.
    • Con A-induced suppressor cells maintained activity in culture with interleukin 2, while untreated LNL did not.

    Conclusions:

    • Suppressor T-cell activity in lymph nodes near melanoma contributes to the observed weak immune reactivity.
    • These findings highlight a mechanism of local immune suppression in the tumor microenvironment.
