Racemic Mixtures and the Resolution of Enantiomers

21.8K
A racemic mixture, or racemate, is an equimolar mixture of enantiomers of a molecule that can be separated using their unique interaction with chiral molecules or media. Racemic mixtures are denoted by the (±)- prefix. This ‘optical rotation descriptor’ applies to the whole solution of a racemic mixture rather than a specific stereoisomer. Enantiomers typically have the same physical and chemical properties. Hence, they are not easily separable. However, enantiomers can exhibit...

Area of Science:

  • Natural Product Chemistry
  • Medicinal Mycology
  • Organic Chemistry

Background:

  • Ganoderma cochlear is a fungus recognized for its edible and medicinal properties.
  • Phytochemical investigations are crucial for discovering novel bioactive compounds from natural sources.

Purpose of the Study:

  • To isolate and characterize new aromatic meroterpenoids from Ganoderma cochlear.
  • To evaluate the antioxidant and anti-proliferative activities of the isolated compounds.

Main Methods:

  • High-Performance Liquid Chromatography with Ultraviolet detection (HPLC-UV) for compound isolation.
  • 1D, 2D Nuclear Magnetic Resonance (NMR) spectroscopy and Mass Spectrometry (MS) for structural elucidation.
  • Electronic Circular Dichroism (ECD) calculations for stereostructure assignment.
  • In vitro assays to determine antioxidant activity and inhibition of hepatic stellate cell proliferation.

Main Results:

  • Nine new aromatic meroterpenoids, cochlearins A-I (1-9), were identified, including six racemates.
  • All isolates exhibited comparable antioxidant activities to trolox, with IC50 values ranging from 3.1 to 5.3 μM.
  • (±)-Cochlearin D (4) and its enantiomer showed weak inhibition of TGF-β1-induced hepatic stellate cell proliferation.

Conclusions:

  • The study successfully identified novel meroterpenoids from Ganoderma cochlear.
  • The isolated compounds possess significant antioxidant potential.
  • Specific compounds show preliminary activity against hepatic stellate cell proliferation, suggesting therapeutic applications.
