Researchers identified novel tregitopes, which are peptide sequences that expand regulatory T cells (Tregs). These tregitopes show potential as immunomodulators for treating autoimmune diseases and in transplantation by promoting immune tolerance.
Area of Science:
- Immunology
- Molecular Biology
- Computational Biology
Background:
- Tregitopes are amino acid sequences that induce regulatory T cell (Treg) expansion.
- Tregs are crucial for immune tolerance, with potential applications in autoimmune diseases and transplantation.
- Previous in silico models guided the selection of potential immunotolerogenic peptides.
Purpose of the Study:
- To select potential tregitopes using an in silico model.
- To assess the impact of selected peptides on Treg expansion in vitro.
- To identify novel tregitopes for immunomodulatory applications.
Main Methods:
- Selected eight peptides based on an in silico model and known immunotolerogenic functions.
- Measured peptide affinity to HLA-DRB1*04:01 using surface plasmon resonance.
- Co-cultured mouse antigen-presenting cells with naive T cells, stimulated with peptides, and analyzed Treg frequency via flow cytometry.
Main Results:
- Two novel tregitopes were identified based on their ability to induce Tregs.
- The identified tregitopes were derived from yeast and adenovirus proteins.
- Peptide affinity to HLA-DRB1*04:01 was assessed as a measure of TCR ligand potential.
Conclusions:
- Novel putative tregitopes were identified, demonstrating potential for Treg induction.
- These tregitopes could serve as immunomodulators in preclinical models.
- The findings support the role of tregitopes in maintaining immune tolerance.
