This study optimized solid lipid nanoparticles (SLNs) to enhance pomegranate extract (PE) anticancer efficacy. The nanoencapsulated PE demonstrated significantly improved anti-proliferative activity and selectivity against cancer cells compared to the free extract.
Area of Science:
- Nanotechnology
- Pharmacology
- Biochemistry
Background:
- Pomegranate extract (PE) possesses natural antioxidant and anticancer properties due to its polyphenol content.
- Maximizing the chemotherapeutic efficacy of PE is a key research objective.
Purpose of the Study:
- To optimize a solid lipid nanoparticle (SLN) formulation for enhanced pomegranate extract (PE) delivery.
- To improve the chemotherapeutic efficacy and selectivity of PE using nanoencapsulation.
Main Methods:
- Utilized Box-Behnken design to optimize lipid, surfactant, and cosurfactant concentrations for PE-loaded SLNs.
- Prepared PE-SLNs via hot homogenization and ultra-sonication.
- Evaluated anti-proliferative activity of optimized PE-SLNs against MCF-7, PC-3, HepG-2 cancer cell lines and HFB-4 normal cells.
Main Results:
- Optimized PE-SLNs achieved particle sizes between 407.5-651.9 nm and entrapment efficiencies of 56.02-65.23%.
- The optimized PE-SLN formulation showed over a 40-fold increase in growth inhibition compared to free PE.
- Demonstrated enhanced selectivity against cancer cells, particularly MCF-7 breast cancer cells, over normal cells.
Conclusions:
- Nanoencapsulation significantly enhances the anticancer efficacy of pomegranate extract.
- Optimized PE-loaded SLNs represent a promising candidate for novel chemotherapeutic applications.
