    Background:

    • Cardiac function and structure are sensitive to oxygen deprivation (anoxia).
    • Age-related changes in cardiac response to stress are not fully understood.
    • Mitochondrial integrity is crucial for cardiac energy metabolism.

    Purpose of the Study:

    • To compare the effects of acute anoxia on left ventricular function and structure in young versus aged male mice.
    • To investigate age-dependent differences in cardiac cellular response to anoxia and reoxygenation.

    Main Methods:

    • Male C57BL/6J mice (3 and 24 months old) were subjected to a 3-minute anoxia exposure.
    • Left ventricular function and structure were assessed before, during, and after anoxia.
    • Cardiac tissue was examined for ultrastructural changes, including mitochondrial morphology and lipid-like vacuoles.

    Main Results:

    • Young mouse hearts exhibited abnormal lipid-like vacuoles, myelin figures, and reduced mitochondrial ATPase during anoxia, leading to irreversible failure upon reoxygenation.
    • Aged mouse hearts maintained normal myofibrillar, mitochondrial, and sarcoplasmic structures throughout anoxia and showed only occasional myelin figures during reoxygenation.
    • Pathological changes were significantly more extensive in young mice compared to aged mice during anoxia and recovery.

    Conclusions:

    • Young hearts are highly susceptible to anoxic injury, with significant structural degradation and functional failure.
    • Older hearts demonstrate remarkable resilience to acute anoxia, preserving structure and function.
    • Age-associated differences in cardiac response to anoxia highlight the protective effects of aging on the myocardium.
