Summary

Three specific microRNAs (miRNAs) were found to be downregulated in individuals with Internet gaming disorder (IGD). These miRNA alterations are associated with a significantly increased risk and may help understand IGD pathophysiology.

Background:

  • Internet gaming disorder (IGD) is a growing concern with potential psychiatric implications.

  • No prior studies have investigated blood miRNA profiles in IGD patients.

Purpose of the Study:

  • To identify specific blood miRNAs associated with Internet gaming disorder (IGD).
  • To explore the diagnostic potential of these miRNAs as biomarkers for IGD.
  • To investigate the underlying neural pathways affected in IGD.

Main Methods:

  • Analyzed miRNA expression profiles in 51 samples (25 IGD, 26 controls) using TaqMan Low Density miRNA Array.
  • Validated findings using quantitative reverse transcription PCR on 36 independent samples (20 IGD, 16 controls).
  • Investigated downstream target gene protein expression via Western blot.

Main Results:

  • Identified three significantly downregulated miRNAs (hsa-miR-200c-3p, hsa-miR-26b-5p, hsa-miR-652-3p) in IGD patients.
  • A higher number of altered miRNAs correlated with increased IGD risk (OR 22).
  • Downstream target genes (GABRB2, DPYSL2) showed significantly higher protein expression in IGD.

Conclusions:

  • Downregulation of hsa-miR-200c-3p, hsa-miR-26b-5p, and hsa-miR-652-3p is observed in Internet gaming disorder (IGD) patients.
  • These miRNAs may serve as potential biomarkers for IGD.
  • Findings contribute to understanding the pathophysiology of IGD.
