Ebselen pretreatment may protect against cisplatin-induced ovarian damage in rats. This antioxidant compound helped preserve ovarian function and reduce tissue injury in a chemotherapy model.
Area of Science:
- Reproductive toxicology
- Pharmacology
- Biochemistry
Background:
- Cisplatin (CP) chemotherapy can cause significant ovarian damage, a major clinical limitation.
- Ovarian damage can lead to reduced fertility and long-term health issues in cancer survivors.
- Ebselen, a seleno-organic compound, possesses known antioxidant and anti-inflammatory properties.
Purpose of the Study:
- To investigate the protective effects of ebselen against cisplatin-induced ovarian damage in a rat model.
- To evaluate the impact of ebselen on oxidative stress markers and ovarian function following cisplatin treatment.
Main Methods:
- Adult female rats were divided into four groups: control, cisplatin (CP) only, CP with saline, and CP with ebselen pretreatment.
- CP was administered intraperitoneally twice weekly for five weeks.
- Ovarian tissue levels of malondialdehyde (MDA), nitric oxide (NOx), glutathione (GSH), Cu/Zn-superoxide dismutase (Cu/Zn-SOD), and catalase were measured.
- Histopathological damage scores (HDSs) and serum antimullerian hormone (AMH) levels were assessed.
Main Results:
- Ebselen pretreatment (Group 4) significantly increased ovarian tissue Cu/Zn-SOD and GSH levels compared to CP-only groups (Groups 2 and 3).
- MDA and NOx levels were significantly reduced in the ebselen-treated group.
- Serum AMH levels and ovarian histopathology were significantly improved in rats pretreated with ebselen.
Conclusions:
- Ebselen demonstrates a protective effect against cisplatin-induced ovarian damage in this experimental model.
- The mechanism of protection appears to involve the mitigation of oxidative stress.
- Ebselen may be a potential therapeutic agent to preserve ovarian function during cisplatin chemotherapy.
