Sperm Transport01:15

Sperm Transport

3.6K
The journey of sperm from its origin to the point of ejaculation begins within the seminiferous tubules of the testis. Here, Sertoli cells produce fluid that propels non-motile sperm through a series of conduits, starting with the straight tubules leading to the rete testis. This interconnected network of tubules acts as the initial pathway for sperm, guiding them into the efferent ductules and then into the epididymis for maturation.
The maturation phase occurs in the epididymis, where sperm...

EDTA: Auxiliary Complexing Reagents01:26

EDTA: Auxiliary Complexing Reagents

1.4K
EDTA titrations are usually carried out in highly basic conditions, where the fully deprotonated form of EDTA, Y4−, actively complexes with the free metal ions in the solution. Several metal ions precipitate as hydrous oxide (hydroxides, oxides, or oxyhydroxides) under these conditions, lowering the concentration of free metal ions in the solution. For this reason, auxiliary complexing agents or ligands such as ammonia, tartrate, citrate, or triethanolamine are used in EDTA titrations to...

Affinity Chromatography01:03

Affinity Chromatography

3.1K
Affinity chromatography is a powerful technique extensively utilized for separating and purifying specific biomolecules from complex mixtures. It capitalizes on the highly selective binding between an analyte and its counterpart, such as antibody-antigen interactions. The counterpart is immobilized on the stationary phase, forming an affinity column. The stationary phase typically consists of solid support, such as agarose or porous glass beads, immobilizing the affinity ligand. The mobile...

Acid Halides to Ketones: Gilman Reagent

4.1K
Lithium dialkyl cuprate, also known as Gilman reagents, selectively reduces acid halides to ketones. The acid chloride is treated with Gilman reagent at −78 °C in the presence of ether solution to produce a ketone in good yield.
As shown below, the mechanism proceeds in two steps. First, one of the alkyl groups of the reagent acts as a nucleophile and attacks the acyl carbon of the acid chloride to form a tetrahedral intermediate. This is followed by the reformation of the carbon–oxygen...

Area of Science:

  • Forensic Science
  • Biotechnology
  • Molecular Biology

Background:

  • Differential extraction is a common method for separating sperm and epithelial cells in forensic samples.
  • Existing methods can be time-consuming and may not efficiently isolate sperm DNA from low-quantity samples.

Purpose of the Study:

  • To demonstrate a proof of concept for affinity-based sperm cell purification using SOMAmer reagents.
  • To evaluate the effectiveness of SOMAmer reagents in separating sperm cells from epithelial cells in mock forensic samples.

Main Methods:

  • SOMAmer reagents were selected in vitro using whole-cell SELEX to specifically bind intact sperm cells.
  • Buccal swabs with added semen were used to test the separation of sperm from epithelial cells and debris.
  • DNA profiles were generated from eluted sperm cells to assess DNA quality.

Main Results:

  • Successful separation of sperm cells from epithelial cells and debris was achieved.
  • Primarily male DNA profiles were generated from sperm cells purified using SOMAmers.
  • The quality of sperm DNA isolated was comparable to existing differential extraction methods, especially at higher sperm concentrations.

Conclusions:

  • SOMAmer reagents can be effectively used for affinity-based purification of sperm cells.
  • This method offers a simple, rapid (<2 hours), and potentially automatable approach for sperm purification.
  • This technique has potential applications in high-throughput sexual assault forensic casework.
