DNA as a Genetic Template02:05

DNA as a Genetic Template

27.9K
Two structural features of the DNA molecule provide a basis for the mechanisms of heredity: the four nucleotide bases and its double-stranded nature. The Watson-Crick model of double-helical DNA structure, proposed in 1952, drew heavily upon the X-ray crystallography work of researchers Rosalind Franklin and Maurice Wilkins. Watson, Crick, and Wilkins jointly received the Nobel Prize in Physiology or Medicine for their work in 1962. Franklin was, controversially, excluded from the prize for...

Feedback Inhibition

57.3K
Biochemical reactions are occurring constantly in cells, converting starting substances to different products, usually with the help of enzymes that speed the reactions. Without enzymes, it would take far too long for most reactions to occur to be useful to the cell!

Calculating the Equilibrium Constant02:46

Calculating the Equilibrium Constant

38.3K
The equilibrium constant for a reaction is calculated from the equilibrium concentrations (or pressures) of its reactants and products. If these concentrations are known, the calculation simply involves their substitution into the Kc expression.
For example, gaseous nitrogen dioxide forms dinitrogen tetroxide according to this equation:

Calculating Standard Free Energy Changes02:49

Calculating Standard Free Energy Changes

25.7K
The free energy change for a reaction that occurs under the standard conditions of 1 bar pressure and at 298 K is called the standard free energy change. Since free energy is a state function, its value depends only on the conditions of the initial and final states of the system. A convenient and common approach to the calculation of free energy changes for physical and chemical reactions is by use of widely available compilations of standard state thermodynamic data. One method involves the...

Calculating pH Changes in a Buffer Solution02:45

Calculating pH Changes in a Buffer Solution

58.9K
A buffer can prevent a sudden drop or increase in the pH of a solution after the addition of a strong acid or base up to its buffering capacity; however, such addition of a strong acid or base does result in the slight pH change of the solution. The small pH change can be calculated by determining the resulting change in the concentration of buffer components, i.e., a weak acid and its conjugate base or vice versa. The concentrations obtained using these stoichiometric calculations can be used...

Summary

A generic template effectively calculates VEMP inhibition depth (VEMPid) for assessing saccular function. This method provides accurate results comparable to subject-specific templates, aiding vestibulopathy diagnosis.

Area of Science:

  • Neuroscience
  • Audiology
  • Vestibular System Research

Background:

  • Cervical vestibular evoked myogenic potentials (cVEMP) assess saccular function via sternocleidomastoid muscle inhibition.
  • VEMP inhibition depth (VEMPid) quantifies saccular inhibition, offering improved accuracy over amplitude normalization, especially at low response levels.
  • Current VEMPid calculation requires a subject-specific template, which is problematic for patients with weak cVEMP responses.

Purpose of the Study:

  • To investigate the efficacy of a "generic" template, derived from healthy subjects, for calculating VEMPid.
  • To determine if a generic template can replace subject-specific templates in VEMPid computation, thereby facilitating clinical application in patients with vestibulopathy.

Main Methods:

  • A generic cVEMP template was created by averaging responses from six healthy individuals.
  • cVEMPs were recorded in 40 healthy subjects using 500, 750, and 1000 Hz tonebursts at varying sound levels (98-123 dB peSPL).
  • VEMPid was calculated using both the generic and subject-specific templates, with performance compared using receiver operating characteristic curves.

Main Results:

  • No significant differences were observed in VEMPid calculations between generic and subject-specific templates across all tested frequencies and sound levels.
  • Receiver operating characteristic curve analysis indicated that both templates performed equally well in determining cVEMP thresholds.
  • The specific shape of the generic template did not significantly impact the accuracy of VEMPid calculations.

Conclusions:

  • A generic template is a viable alternative to subject-specific templates for calculating VEMPid.
  • VEMPid offers advantages over traditional amplitude-normalized cVEMPs, including a baseline of zero with no stimulus and real-time data acquisition feedback.
  • The use of a generic template simplifies VEMPid application, potentially improving the clinical assessment of vestibulopathy.
