Hormones—or any molecule that binds to a receptor, known as a ligand—that are lipid-insoluble (water-soluble) are not able to diffuse across the cell membrane. In order to be able to affect a cell without entering it, these hormones bind to receptors on the cell membrane.   • Murine islets were cultured in vitro with either silk fibroin or alginate.

Main Results:

  • While inflammatory cytokines reduced GSIS in all groups, islets preconditioned with silk fibroin showed better preservation of function compared to alginate or medium alone.
  • Silk fibroin preconditioning significantly increased GLUT2 transporter expression on islet cell surfaces.
  • This increased GLUT2 expression persisted even under inflammatory cytokine stress.

Conclusions:

  • Silk fibroin preconditioning enhances islet cell surface GLUT2 expression.
  • This effect may contribute to improved islet engraftment and function following transplantation, particularly under inflammatory stress.
  • Silk fibroin represents a promising biomaterial for improving islet transplantation outcomes.
