Background:

  • Natural killer (NK) cells are crucial for innate immunity and cancer surveillance.
  • Efficient methods for expanding NK cells are needed for therapeutic applications.

Purpose of the Study:

  • To develop an easy and effective method for amplifying NK cells from peripheral blood mononuclear cells (PBMNCs) in vitro.
  • To establish a foundation for NK cell-based immunotherapies.

Main Methods:

  • PBMNCs were isolated from umbilical cord blood and cultured for 14 days.
  • Cultures were stimulated with anti-CD16 monoclonal antibody (mAb), anti-CD3 mAb, or both, in serum-free media with IL-2, IL-15, and IL-21.
  • Cell proliferation, NK cell purity (CD56+CD3-), and cytotoxic activity against K562 target cells were assessed.

Main Results:

  • Culture with anti-CD3 mAb resulted in significantly higher total cell expansion compared to anti-CD16 mAb or combined stimulation.
  • High purity of NK cells (approximately 80%) was achieved across all culture conditions.
  • NK cell cytotoxicity remained comparable among groups at various effector/target ratios.

Conclusions:

  • Incubation with anti-CD3 monoclonal antibody, IL-2, IL-15, and IL-21 effectively expands highly purified NK cells from mononuclear cells.
  • This method offers a simple and promising approach for generating NK cells for therapeutic purposes.
