Insights

Ataxia Telangiectasia and Rad3 related protein (ATR) inhibitors NVP-BEZ235 and AZD6738 show good brain penetration for glioblastoma treatment. However, neither drug improved survival when combined with radiation, suggesting they are not effective radiosensitizers.

Area of Science:

  • Oncology
  • Pharmacology
  • Radiotherapy

Background:

  • Ataxia Telangiectasia and Rad3 related protein (ATR) is crucial in DNA damage response, potentially causing cancer cell resistance to radiotherapy.
  • NVP-BEZ235 and AZD6738 are ATR inhibitors proposed as radiosensitizers for glioblastoma (GB).

Purpose of the Study:

  • To evaluate the brain penetration of NVP-BEZ235 and AZD6738.
  • To assess the radiosensitizing potential of these ATR inhibitors in glioblastoma models.

Main Methods:

  • Mice received NVP-BEZ235 or AZD6738 intraperitoneally.
  • Blood and brain samples were collected at various time points to measure drug concentrations using High Performance Liquid Chromatography/Mass Spectrometry.
  • Tumor-bearing mice were treated with drugs and ionizing radiation to assess toxicity and survival.

Main Results:

  • Both NVP-BEZ235 and AZD6738 demonstrated effective brain penetration and bioavailability after systemic administration.
  • A trend towards toxicity was observed with NVP-BEZ235 plus ionizing radiation compared to AZD6738 plus radiation or vehicle plus radiation.
  • No significant improvement in survival was observed for any treatment group.

Conclusions:

  • NVP-BEZ235 and AZD6738 exhibit favorable pharmacokinetic properties for brain penetration.
  • Despite promising pharmacokinetics, these ATR inhibitors do not show efficacy as radiosensitizers for glioblastoma in this study.

Protein Kinases and Phosphatases

Proteins undergo chemical modifications that trigger changes in the charge, structure, and conformation of the proteins. Phosphorylation, acetylation, glycosylation, nitrosylation, ubiquitination, lipidation, methylation, and proteolysis are various protein modifications that regulate protein activity. Such modifications are usually enzyme-driven.

Many proteins in the cell are regulated by phosphorylation, the addition of a phosphate group. A family of enzymes called kinases...

Functional Brain Systems: Limbic System

The limbic system, often called the "emotional brain," is a complex set of structures located deep within the brain. The intricate network of the limbic system supports a wide range of psychological functions, from emotional regulation to memory formation and sensory processing. This functional brain region encompasses specific parts of the diencephalon and the cerebrum, integrating the higher mental functions of the cerebral cortex with the primitive emotional responses of the deep brain...

Treatment for Pulmonary Arterial Hypertension: Receptor Tyrosine Kinase Inhibitors and Calcium Channel Blockers

Receptor tyrosine kinase inhibitors (TKIs) and calcium channel blockers (CCBs) are two critical categories of drugs employed in the treatment of pulmonary artery hypertension (PAH). PAH is a disease that causes high blood pressure in the pulmonary arteries, resulting in chest pain, fatigue, and shortness of breath.
TKIs, such as imatinib (Gleevec), are particularly effective in tackling the growth and mitogenic factors that become upregulated in PAH patients. These factors contribute to the...

Functional Brain Systems: Reticular Formation

The reticular formation is a complex network of gray and white matter located within the brainstem extending from the medulla to the midbrain.
Within the reticular formation, there are several distinct nuclei that can be classified into three broad categories. The Raphe nuclei are located along the midline of the brainstem. They are primarily known for their role in synthesizing and releasing serotonin, a neurotransmitter involved in regulating mood, appetite, sleep, and circadian rhythms. The...

Eukaryotic Transcription Inhibitors01:52

Eukaryotic Transcription Inhibitors

Certain biochemical processes, such as embryonic development and cell growth regulation, depend on the repression of specific genes. DNA binding proteins known as eukaryotic transcription inhibitors regulate the repression of gene expression in eukaryotes. The presence of these inhibitors at the required location and time in the cell is triggered by the presence of hormones and additional signals from other cells.
Eukaryotic transcription inhibitors usually contain two distinct domains, a...
