Background:

Migraine trials use pain relief and sustained pain relief as key efficacy measures.
Existing methods for estimating sustained pain relief with missing data are statistically inconsistent.
Methods for non-ignorable missing data require untestable assumptions about the missing data mechanism.

Purpose of the Study:

To propose two novel, statistically consistent methods for estimating sustained pain relief in migraine trials with missing data.
To avoid complex modeling assumptions regarding missing data mechanisms and correlation structures.

Main Methods:

Development of two intuitive imputation-based methods tailored to the definition of sustained pain relief.
Theoretical consistency analysis of the proposed methods.
Extensive simulation studies to evaluate empirical performance.

The proposed methods demonstrate good performance in reducing bias and mean square error in simulations.
The methods are robust except in rare, extreme missing data scenarios.
The methods were successfully applied to a real migraine trial dataset.
