Background:

  • Neurotransmitter deficits (dopamine, acetylcholine, etc.) are observed in Alzheimer's disease (AD) brains.
  • The hypothalamus, crucial for neuroendocrine regulation, exhibits pathological changes in AD.
  • Despite these indicators, neuroendocrine function in AD patients remains under-investigated.

Purpose of the Study:

  • To compare neuroendocrine function in patients with probable AD versus non-demented controls.
  • To investigate basal and stimulated plasma levels of key pituitary hormones and endorphins.

Main Methods:

  • Radioimmunoassays (RIA) were used to measure plasma levels of prolactin (PRL), growth hormone (GH), and thyroid-stimulating hormone (TSH).
  • Hormone levels were assessed before and after stimulation with metoclopramide, l-dopa, or thyrotropin-releasing hormone.

  • Basal plasma levels of beta-endorphin and beta-lipotropin were measured using RIA post-high-performance liquid chromatography.
  • Main Results:

    • Basal and stimulated plasma levels of PRL, GH, TSH, and beta-lipotropin were comparable between AD patients and controls.

    • This elevation in beta-endorphin may suggest decreased tuberoinfundibular dopaminergic activity, a finding also noted in Parkinson's disease.

    Conclusions:

    • Neuroendocrine dysfunction, specifically elevated beta-endorphin, may be present in Alzheimer's disease.
    • While most pituitary hormone responses were similar, the beta-endorphin finding warrants further investigation for clinical significance.
    • The results suggest potential links between dopaminergic activity and neuroendocrine alterations in AD.
