This study presents a novel method for gene silencing using self-assembled multilayers of small interfering RNA (siRNA) and poly-L-lysine (PLL). These siRNA/PLL multilayers demonstrate efficient gene silencing for tissue engineering applications without significant cytotoxicity.
Area of Science:
- Biomaterials Science
- Molecular Biology
- Tissue Engineering
Background:
- Small interfering RNA (siRNA) is crucial for controlling cellular functions in tissue engineering.
- Traditional polycationic carriers for siRNA face challenges due to low siRNA association and cytotoxicity.
Purpose of the Study:
- To develop an efficient and less cytotoxic method for siRNA delivery.
- To explore the self-assembly of siRNA with mild polyelectrolytes for gene silencing.
Main Methods:
- Fabrication of siRNA/poly-L-lysine (PLL) multilayers via layer-by-layer deposition on a polydopamine-coated substrate.
- Assessment of gene silencing efficiency in cells cultured on the fabricated multilayers.
- Investigation of the impact of multilayer structure on gene silencing efficacy.
Main Results:
- The siRNA/PLL multilayers effectively inhibited target gene expression in cells.
- Gene silencing efficiency was correlated with the number of siRNA layers in the multilayer.
- This approach offers a promising alternative to conventional siRNA delivery methods, reducing cytotoxicity.
Conclusions:
- Self-assembled siRNA/PLL multilayers provide an efficient platform for surface-mediated gene silencing.
- This technique holds potential for advanced tissue engineering applications requiring controlled gene modulation.

Stratum Basale
Stratum basale, also known as the stratum germinativum, is the deepest layer of the epidermis. It is composed of a single layer of actively dividing cells called basal cells or basal keratinocytes. These cells constantly undergo cell division to replenish the upper layers of the epidermis. Additionally, melanocytes, which...
