Background:

  • Previous pulmonary artery banding models for right ventricular hypertrophy (RVH) in adult animals suffer from unpredictable responses and high mortality.
  • A need exists for a more reliable and reproducible method to induce RVH in animal models.

Purpose of the Study:

  • To evaluate the efficacy of progressive pulmonary artery banding in young pigs for creating a reliable model of right ventricular hypertrophy (RVH).

Main Methods:

  • Sixteen 6-week-old Yorkshire pigs underwent placement of a nonconstricting Dacron strip around the pulmonary artery (PA) via left minithoracotomy.
  • Animals were studied after 2-3 months of growth, with five sham-operated pigs serving as controls.
  • Measurements included right ventricular free wall (RVFW) to total heart weight ratio, RVFW to left ventricular free wall (LVFW) weight, and septal ratio.

  • The pulmonary artery banding group showed significantly increased RVFW to total heart weight ratio (0.38 vs. 0.28) and RVFW to LVFW weight (1.09 vs. 0.66).
  • A decrease in LVFW to total heart weight ratio (0.36 vs. 0.45) was observed, with no significant change in the septal ratio, indicating concomitant septal hypertrophy.
  • No mortality occurred during the growth period in the banded group.

Conclusions:

  • This method offers a low-mortality model for studying RVH during maturation.
