MicroRNA values in children with rheumatic carditis: a preliminary study

Purpose of the Study:

• To investigate and compare miRNA expression levels in children diagnosed with rheumatic carditis against healthy controls.
• To identify potential miRNA biomarkers for the early detection or monitoring of rheumatic carditis in pediatric patients.

Main Methods:

• Real-time polymerase chain reaction (PCR) was employed to analyze the expression of 20 specific miRNAs in 36 children with rheumatic carditis and 35 healthy controls.
• Conventional echocardiography was performed on all participants to assess cardiac function.
• Expression levels of miRNAs including hsa-miR-16-5p, hsa-miR-223-3p, and hsa-miR-92a-3p were quantified and compared between the patient and control groups.

• A significant downregulation of hsa-miR-16-5p (1.46-fold decrease, p < 0.01) and hsa-miR-223-3p (1.46-fold decrease, p < 0.01) was observed in children with rheumatic carditis.
• hsa-miR-92a-3p expression was also significantly lower (1.27-fold decrease, p < 0.05) in the rheumatic carditis group compared to controls.
• No significant differential expression was found for the other 17 examined miRNAs between the two groups.

Conclusions:

• The study identifies a significant downregulation of hsa-miR-16-5p, hsa-miR-223-3p, and hsa-miR-92a-3p in children suffering from rheumatic carditis.
• These specific miRNAs show potential as novel biomarkers for rheumatic carditis in pediatric populations.
• Further research is warranted to validate these findings and explore the clinical utility of these miRNAs as diagnostic or prognostic markers.
