Purpose of the Study:

  • To evaluate the effectiveness of direct PCR for STR DNA profiling from various touch DNA samples relevant to forensic investigations.
  • To compare the performance of Identifiler® Plus and GlobalFiler® kits in direct PCR amplification of touch DNA.

Main Methods:

  • Volunteers handled eight different sample types for up to 15 seconds to deposit trace DNA.
  • Direct PCR was performed on these samples using Identifiler® Plus or GlobalFiler® kits without increasing cycle numbers.
  • A comparison was made between direct PCR and standard DNA extraction using Identifiler®.

Main Results:

  • Informative STR profiles were obtained from all eight substrates using both kits.
  • Identifiler® Plus achieved a 96% allele amplification rate, significantly higher than GlobalFiler®'s 60%.
  • Direct PCR yielded 100% informative profiles, compared to only 4% with standard extraction.

Conclusions:

  • Direct PCR is a highly effective, fast, and non-destructive method for enhancing STR DNA profiles from touch DNA.
  • Identifiler® Plus demonstrated superior performance over GlobalFiler® for direct PCR amplification of touch DNA.
  • Direct PCR offers a viable alternative for generating crucial genetic data from challenging forensic samples where standard methods fail.
