    Background:

    • Polymorphonuclear leukocytes (PMN) play a key role in fighting bacterial infections.

    Purpose of the Study:

    • To investigate the phagocytosis and killing capacity of PMN in preterm and term newborn infants.
    • To compare the function of PMN from newborns to that of adults.
    • To identify potential immunological deficits in neonatal PMN.

    Main Methods:

    • Isolated PMN from 2 ml of venous blood from preterm infants (n=10), term newborn infants (n=9), and adults (n=14).
    • Assessed phagocytosis and killing of staphylococci using isolated PMN.
    • Measured superoxide anion production and activation times in response to phorbolmyristate acetate stimulation.

    Main Results:

    • PMN phagocytosis was comparable across preterm infants, term newborns, and adults.
    • PMN killing capacity was significantly impaired in preterm infants compared to term newborns and adults.
    • Superoxide anion production and activation times were similar in PMN from all groups.

    Conclusions:

    • PMN from term newborn infants exhibit normal phagocytotic and bactericidal capacity.
    • PMN from preterm newborn infants demonstrate diminished bactericidal capacity.
    • The findings suggest impaired bacterial defense mechanisms in preterm neonates, similar to stressed newborns.
