Summary

Multiple polydioxanone (PDO) threads significantly increased collagen production compared to single strands in a rat model. Collagen levels peaked at two weeks and declined by twelve weeks, with retained threads forming fibrous capsules.

Purpose of the Study:

  • To evaluate the impact of varying numbers of polydioxanone (PDO) threads on collagen production and tissue response.
  • To compare PDO threads with poly-lactic acid (PLA) threads in a preclinical model.

Main Methods:

  • Rats received different configurations of PDO threads and monofilament PLA threads in dorsal skin compartments.
  • Tissue samples were analyzed at 1, 2, and 12 weeks for collagen quantification (real-time PCR) and histopathology.

Main Results:

  • Multiple PDO threads yielded higher collagen levels at 2 weeks compared to single strands.
  • Single-stranded PLA threads showed increased collagen type 1 alpha 1 (Col1α1) and collagen type 1 alpha 3 (Col1α3) at week 2.
  • Collagen levels significantly decreased by week 12, with observed fibrous encapsulation of retained threads.

Conclusions:

  • Increasing the number of PDO threads enhances collagen synthesis.
  • PDO threads show potential for stimulating collagen, but long-term effects and degradation patterns require further investigation.
