Differences in methylation on the active and inactive human X chromosomes.

S Lindsay, M Monk, R Holliday

    Annals of Human Genetics
    |May 1, 1985
    PubMed
    Summary

Methylation patterns on the inactive X chromosome were studied. Differentially methylated sites were found, but they do not appear to maintain X chromosome inactivation in human cells.

    Area of Science:

    • Genetics
    • Epigenetics
    • Human X Chromosome Biology

    Background:

    • X chromosome inactivation is a crucial process in female mammals.
    • DNA methylation is a key epigenetic mechanism involved in gene regulation.

    Purpose of the Study:

    • To investigate CCGG site methylation patterns in specific X chromosome regions.
    • To determine the role of differential methylation in maintaining X chromosome inactivation.

    Main Methods:

    • Analysis of DNA methylation using X-chromosome clones from human peripheral blood cells.
    • Comparison of methylation patterns between active and inactive X chromosomes.

    Main Results:

    • Unmethylated CCGG sites correlated with the inactive X chromosome near two random clones and the PGK gene.

    Related Experiment Videos

  • One random clone showed unchanged methylation between active and inactive X chromosomes.
  • Both active and inactive X chromosomes were undermethylated in lymphoblastoid cells, suggesting no functional role for observed differential methylation.
  • Conclusions:

    • Differential methylation at specific CCGG sites on the X chromosome does not appear to be functionally involved in maintaining X chromosome inactivation.
    • Further research is needed to understand the complete epigenetic landscape of X chromosome inactivation.
