Abstract

Insights

Losartan and nitro-oleic acid show potential in preventing and treating diabetic kidney disease by reducing parathyroid hormone-related protein (PTHrP) and improving kidney function and structure.

Area of Science:

  • Nephrology
  • Endocrinology
  • Pharmacology

Background:

  • Parathyroid hormone-related protein (PTHrP) is implicated in angiotensin II (AngII)-induced renal injury.
  • Diabetic nephropathy (DN) is a significant complication of diabetes mellitus.

Purpose of the Study:

  • To compare the preventive and therapeutic effects of AngII antagonists, losartan and nitro-oleic acid (NO2-OA), on diabetic nephropathy in rats.
  • To evaluate the impact of these agents on PTHrP modulation and kidney parameters.

Main Methods:

  • Adult male Sprague Dawley rats were divided into control, DN, and treatment groups (pre- and post-DN with losartan or NO2-OA).
  • Evaluated systolic blood pressure, glucose, GFR, urea, albuminuria, AngII, and renal PTHrP gene expression.
  • Histopathological analysis included Masson's trichrome, PAS staining, and scoring of tubular and glomerular damage.

Main Results:

  • Prophylactic losartan and NO2-OA improved blood pressure, glucose control, kidney function, and reduced AngII and PTHrP levels.
  • Losartan and NO2-OA treatments generally improved kidney parameters, though post-DN NO2-OA had less effect on kidney function.
  • Strong correlations were found between PTHrP and SBP, glucose, AngII, and kidney function. Histopathology showed improvements, particularly with prophylactic losartan.

Conclusions:

  • Both pre- and post-DN administration of losartan and NO2-OA may protect against and help regress diabetic nephropathy.
  • These effects are potentially mediated by the reduction of PTHrP overexpression.

Hormonal Regulation01:40

Hormonal Regulation

Hormones regulate a significant portion of digestion through activation of the neuroendocrine system. The neuroendocrine system of digestion contains many different hormones all with multiple functions that are both, directly and indirectly, involved in digestion.

Hormonal Regulation01:33

Hormonal Regulation

The renin-aldosterone system is an endocrine system which guides the renal absorption of water and electrolytes, thus managing blood pressure and osmoregulation. Activation of the system begins in the kidneys with a small cluster of cells adjacent to the afferent and efferent blood vessels of the renal corpuscle. As the nephrons are filtering blood, juxtaglomerular cells monitor blood pressure. If they detect a decrease in pressure, they release the hormone renin into the bloodstream.

Types of Hormones

Hormones can be classified into three main types based on their chemical structures: steroids, peptides, and amines. Their actions are mediated by the specific receptors they bind to on target cells.

Types of Hormones

Hormones are classified into four main groups: steroids, eicosanoids, amino acid-based derivatives, and peptide hormones.
Steroids and eicosanoids fall under the category of lipid-soluble hormones. Steroids are derived from cholesterol and feature four interconnected carbon rings with variable side chains. Notable examples include estradiol from ovaries and testosterone from testes, exemplifying the critical roles of these lipid-soluble hormones in reproductive physiology. Eicosanoids, derived...

Plant Hormones01:56

Plant Hormones

Plant hormones—or phytohormones—are chemical molecules that modulate one or more physiological processes of a plant. In animals, hormones are often produced in specific glands and circulated via the circulatory system. However, plants lack hormone-producing glands.

The Parathyroid Glands00:59

The Parathyroid Glands

The two pairs of parathyroid glands embedded within the posterior surface of the thyroid gland are restricted by a dense capsule around them. These glands comprise two distinct cell populations—parathyroid oxyphil and parathyroid principal cells- pivotal in calcium homeostasis.
Oxyphil cells, whose functions remain elusive, emerge during late puberty, adding a layer of complexity to the parathyroid gland's intricacies. In contrast, principal parathyroid cells undertake a vital role by...
