    Specific B cells efficiently present tetanus toxoid (TT) to T cells at very low concentrations. This antigen presentation requires antigen internalization, processing, and surface display for T cell recognition.

    Background:

    • Understanding the antigen bridge between B lymphocytes and T lymphocytes is crucial for immune response.
    • Tetanus toxoid (TT) is a well-characterized antigen used to study specific immune responses.

    Purpose of the Study:

    • To investigate the efficiency of antigen presentation by specific B cells to T cells.
    • To elucidate the mechanism of antigen uptake, processing, and presentation by B cells.

    Main Methods:

    • Isolation of human T cell clones and EBV-transformed B cell clones specific for tetanus toxoid (TT).
    • Assays for T cell proliferation in response to antigen presented by B cells.
    • Experiments using fixed and chloroquine-treated B cells to study antigen processing.
    • Inhibition assays using antibodies against T4 antigen.

    Main Results:

    • TT-specific B cells presented TT with extremely high efficiency, triggering T cell proliferation at 10(-12) M TT.
    • This concentration is 10,000 times lower than required for conventional antigen-presenting cells.
    • Antigen presentation was MHC-restricted and required antigen internalization, processing, and surface display.
    • Surface antibodies were mainly involved in antigen uptake, not presentation itself.

    Conclusions:

    • Specific B cells are highly potent antigen-presenting cells for cognate T cells.
    • The interaction between specific B and T cells exhibits high avidity.
    • Antigen processing and presentation by B cells is a multi-step process involving internalization and surface display.
