Immunotherapy is a treatment that boosts or manipulates the immune system to fight diseases, including cancer. For instance, by stimulating an immune response through vaccinations against viruses that cause cancers, like hepatitis B virus and human papillomavirus, these diseases can be prevented. Nonetheless, some cancer cells can avoid the immune system due to their rapid mutation and division. The immune response to many cancers involves three phases: elimination, equilibrium, and escape.

Literature search of PubMed, Scopus, and Google Scholar.
Selection of recent randomized double-blinded placebo-controlled trials and meta-analyses.
Focus on current and novel immunotherapy approaches.

Main Results:

Novel adjuvants (e.g., TLR-4 agonists), nanoparticles, and virus-like particles show promise.
Intralymphatic, epicutaneous, and modified allergen strategies demonstrate efficacy and shortened courses.
Omalizumab reduces side effects but not efficacy; conventional extracts remain standard for long-term tolerance.

Conclusions:

New immunotherapy routes, modified allergens, and adjuvants aim to enhance immune tolerance.
Phase 3 and head-to-head trials are needed to confirm efficacy and safety of novel approaches.
