    Background:

    • Ethanol consumption can affect glucose homeostasis and insulin secretion.
    • Adrenergic receptors and cyclic AMP (cAMP) are known regulators of insulin release.

    Purpose of the Study:

    • To investigate the role of alpha- and beta-adrenergic receptors and cyclic AMP in mediating ethanol's effects on insulin release from isolated rat islets.

    Main Methods:

    • Isolated rat islets were treated with alpha-adrenergic blocker (phentolamine) or beta-adrenergic blocker (propranolol).
    • Ethanol (1 g/kg) was administered intragastrically prior to sacrifice.
    • Insulin release was measured in response to glucose, dibutyryl-cyclic AMP, and theophylline.

    Main Results:

    • Phentolamine enhanced glucose-mediated insulin release, while propranolol decreased it.
    • Ethanol inhibited glucose-induced insulin release in all treatment groups.
    • Ethanol attenuated insulin release stimulated by dibutyryl-cyclic AMP and inhibited theophylline-enhanced insulin release.

    Conclusions:

    • Ethanol's detrimental effects on insulin release from isolated rat islets do not appear to involve the adrenergic system.
    • The mechanism of ethanol's inhibition of insulin release does not directly involve the cyclic AMP pathway.
