Summary

New multiple-locus variable number of tandem repeats analysis (MLVA) and tandem repeat sequence typing (TRST) methods effectively subtype Staphylococcus lugdunensis infections. These molecular tools offer excellent discriminatory power for tracking bacterial strains in epidemiology.

  • To develop and evaluate novel molecular typing methods for S. lugdunensis.
  • To compare the discriminatory power of new methods against existing techniques.

Main Methods:

  • Development of multiple-locus variable number of tandem repeats analysis (MLVA) and tandem repeat sequence typing (TRST) methods.
  • Testing of 128 S. lugdunensis isolates using MLVA, TRST, multilocus sequence typing (MLST), and multivirulence-locus sequence typing (MVLST).

Main Results:

  • MLVA and TRST demonstrated excellent typeability, reproducibility, and epidemiological concordance.
  • The new methods showed higher discriminatory power than MLST and MVLST.
  • Clustering analysis revealed no association between bacterial lineages and clinical settings.

Conclusions:

  • MLVA and TRST are valuable tools for molecular epidemiological studies of S. lugdunensis.
  • These methods provide superior strain differentiation within clonal lineages of S. lugdunensis.
