Flow cytometry in brain tumors. I. Ploidy abnormalities.

K Christov, Z Zapryanov

    Summary

Flow cytometry (FCM) estimated DNA content in 75 brain tumors, finding aneuploid cell populations in most high-grade astrocytomas. This DNA analysis aids in understanding tumor malignancy and progression.

    Area of Science:

    • Neuro-oncology
    • Cytometry
    • Molecular Pathology

    Background:

    • Flow cytometry (FCM) is a valuable technique for analyzing cellular DNA content.
    • Brain tumors, including astrocytomas and meningiomas, exhibit diverse genetic profiles.
    • Accurate tumor grading is crucial for determining prognosis and treatment strategies.

    Purpose of the Study:

    • To evaluate the utility of flow cytometry for DNA content estimation in a cohort of 75 brain tumors.
    • To correlate DNA aneuploidy patterns with histological grading of astrocytomas.
    • To investigate the prevalence of aneuploid cell populations in different brain tumor types.

    Main Methods:

    • Flow cytometry was employed to measure the DNA content of cells from 75 surgically resected brain tumors.
    • Tumor samples included astrocytomas (38 cases) and meningiomas (16 cases), among others.
    • Astrocytomas were histologically classified into three grades of malignancy.

    Main Results:

    • Aneuploid cell populations were detected in 1/4 of grade I astrocytomas, 11/20 of grade II astrocytomas, and 10/14 of grade III astrocytomas.
    • The DNA index (DI) for most aneuploid astrocytomas ranged between 1 and 2.
    • Multiple aneuploid cell populations were observed in 36% of all aneuploid tumors analyzed.

    Conclusions:

    • Flow cytometry is effective for detecting DNA aneuploidy in brain tumors, particularly astrocytomas.
    • The presence and degree of DNA aneuploidy correlate with the histological malignancy grade of astrocytomas.
    • FCM-based DNA content analysis provides valuable prognostic information for brain tumor patients.
