Insights

Genetic variants in Glutathione S-transferase M1 (GSTM1) and Apolipoprotein E (ApoE) are linked to diastolic dysfunction in beta-thalassaemia patients. These findings highlight genetic predispositions to cardiac injury from iron overload.

Area of Science:

  • Cardiology
  • Genetics
  • Hematology

Background:

  • Cardiomyopathy is a complication of iron loading in beta-thalassaemia patients due to blood transfusions.
  • Excess iron generates free radicals, leading to oxidative damage.
  • Apolipoprotein E (ApoE) and Glutathione S-transferase (GST) are key in scavenging these free radicals.

Purpose of the Study:

  • To investigate the association between genetic variants of ApoE and GST and diastolic dysfunction in beta-thalassaemia patients.
  • To understand the role of these genetic variants in oxidative stress-induced cardiomyopathy.

Main Methods:

  • Echocardiography was performed on 105 beta-thalassaemia patients (>10 years old).
  • Genotyping of ApoE and GST variants was done using PCR-RFLP.
  • Serum GST levels were measured using ELISA.

Main Results:

  • Diastolic dysfunction was present in 22.8% of patients; left ventricular hypertrophy in 35.2%.
  • A significant association was found between the GSTM1 null allele and diastolic dysfunction.
  • The ApoE E2 allele also showed a positive association with diastolic dysfunction. Serum GST levels correlated positively with diastolic function parameters (e/a and e/e' ratios).

Conclusions:

  • The Glutathione S-transferase M1 null allele and Apolipoprotein E E2 allele predispose individuals to cardiac injury.
  • These genetic factors may contribute to oxidative stress-induced heart damage in beta-thalassaemia.

Histone Variants at the Centromere

Histone variants are the histone proteins with structural and sequence variations. These variants may be regarded as “mutant” forms that replace their canonical histone counterparts in the nucleosomes. Specific post-translational modifications on the histone variants enable further chromatin complexity and regulate tissue-specific gene expression. The most common histone variants are from histone H2A, H2B, and linker histone H1 families. However, several variants of histone H3...

Major Organs of the Digestive System

The digestive system is responsible for the ingestion of food, secretion of enzymes, mixing and digesting food, absorption of the nutrients and defecation. The human digestive system consists of two major parts: the gastrointestinal tract and the accessory digestive organs.

Major Hormones and Their Functions01:27

Major Hormones and Their Functions

Hormones, the biochemical messengers produced by endocrine glands, are pivotal in regulating bodily functions and maintaining homeostasis. Each hormone's balance is crucial; imbalances can lead to significant physiological disruptions. Major hormones include oxytocin, cortisol, epinephrine, estrogen, testosterone, thyroxine, growth hormone, insulin, and glucagon.
Oxytocin, produced in the hypothalamus and released by the pituitary gland, plays a role in social bonding, childbirth, and...

Major Losses in Pipes01:28

Major Losses in Pipes

When a fluid flows through a pipe, it experiences energy losses due to frictional resistance along the pipe walls, known as major losses. These energy losses result in a pressure drop, which varies based on the flow conditions — whether laminar or turbulent — and the specific physical properties of the fluid and pipe.
Fluid flow can be classified as laminar or turbulent, primarily based on the Reynolds number. This dimensionless number reflects the relative influence of inertial to viscous...

Association Areas of the Cortex01:21

Association Areas of the Cortex

Association areas are regions of the cerebral cortex that do not have a specific sensory or motor function. Instead, they integrate and interpret information from various sources to enable higher cognitive processes such as memory, learning, and decision-making. Some key association areas include the following:
Prefrontal Association Area: This area is located in the frontal lobe and is involved in planning, decision-making, and moderating social behavior. It connects with primary motor areas,...

Associative Learning01:27

Associative Learning

Associative learning is a fundamental concept in behavioral psychology, wherein a connection is established between two stimuli or events, leading to a learned response. This process is critical in understanding how behaviors are acquired and modified. Conditioning, the mechanism through which associations are formed, can be divided into two main types: classical conditioning and operant conditioning, each elucidating different aspects of associative learning.
Classical conditioning, also known...
