Asiaticoside counteracts the in vitro activation of microglia and astrocytes: Innuendo for multiple sclerosis
1Department of Pharmacology, Acharya & BM Reddy College of Pharmacy, Bengaluru, India.
Background:
During the development of Multiple Sclerosis (MS) there is a marked activation of microglia and astrocyte, leading to progressive inflammation and degeneration of myelin sheath which results in axonal loss and neuron damage.
Purpose:
In this study, we have explored the action of Asiaticoside A against the activated microglia and astrocytes.
Methods:
Primary microglia and astrocyte cultures were used for the study and the activity were evaluated using cell proliferation assay, nitrate assay and TNFα estimation using ELISA.
Results:
Asiaticoside A inhibited the production of nitric oxide and TNFα in LPS activated primary microglia and astrocyte cultures.
Conclusion:
This study suggests that Asiaticoside A may be effective against the progression of MS.
Insights
Asiaticoside A reduces inflammation in brain cells, showing potential to slow Multiple Sclerosis (MS) progression. This compound targets activated microglia and astrocytes, key players in MS.
Area of Science:
- Neuroscience
- Immunology
- Pharmacology
Background:
- Multiple Sclerosis (MS) involves microglia and astrocyte activation, causing inflammation and neurodegeneration.
- This neuroinflammation leads to myelin sheath damage, axonal loss, and neuron injury.
Purpose of the Study:
- To investigate the therapeutic potential of Asiaticoside A.
- To evaluate the effects of Asiaticoside A on activated microglia and astrocytes.
Main Methods:
- Primary microglia and astrocyte cultures were utilized.
- Cell proliferation, nitric oxide production, and TNFα levels were measured.
- Enzyme-linked immunosorbent assay (ELISA) was employed for TNFα quantification.
Main Results:
- Asiaticoside A significantly inhibited nitric oxide production in activated microglia and astrocyte cultures.
- Tumor Necrosis Factor-alpha (TNFα) production was also suppressed by Asiaticoside A.
- These findings indicate an anti-inflammatory effect of Asiaticoside A.
Conclusions:
- Asiaticoside A demonstrates neuroprotective properties by reducing inflammation.
- The compound shows promise as a potential therapeutic agent for Multiple Sclerosis.
- Further research is warranted to explore Asiaticoside A's efficacy in MS treatment.

Multiple Regression
Farmers can use multiple regression to determine the crop yield based on more than one factor, such as water availability, fertilizer, soil properties, etc. Here, the crop yield is the response or dependent variable as it depends on the other independent variables. The analysis requires the construction of a scatter plot...
Multiple Voltage Sources
In series, the positive terminal of one battery is connected to the negative terminal of another battery. Hence, the voltage of each battery is added to give the net voltage, which is increased because each battery boosts the electrons that enter it. The same current flows through each battery because they are connected in series.
Batteries are...
