    Background:

    • Accurate histopathologic diagnosis of thyroid neoplasms is crucial.
    • Enzyme histochemistry offers potential markers for differentiating benign and malignant thyroid tissues.

    Purpose of the Study:

    • To identify enzyme histochemical markers for the diagnosis of human thyroid neoplasms.
    • To evaluate the utility of adenosine triphosphatase (ATP) staining in distinguishing thyroid tumors.

    Main Methods:

    • Surgically removed human thyroid tissue specimens were fixed and embedded.
    • Thin sections were analyzed using enzyme histochemical staining for adenosine triphosphatase, alkaline and acid phosphatases, alpha-naphthyl acetate esterase, and 5'-nucleotidase.
    • Cytologic and enzyme preservation were assessed.

    Main Results:

    • Thyroid epithelial cells and benign neoplasms were positive for 5'-nucleotidase, alpha-naphthyl acetate esterase, and acid phosphatase.
    • Benign tissues were typically negative for adenosine triphosphatase and alkaline phosphatase.
    • Papillary and follicular carcinomas showed adenosine triphosphatase staining, which was also observed in benign glands under specific conditions like Graves' disease.

    Conclusions:

    • Adenosine triphosphatase staining appears to be a valuable adjunct in distinguishing between benign and malignant neoplasms of the thyroid.
    • Enzyme histochemistry, particularly ATP staining, can aid in the histopathologic diagnosis of thyroid neoplasms.
