    Background:

    • Dioscorea nipponica polysaccharides (DNPs) are valuable bioactive compounds with numerous health benefits.
    • Effective purification of DNPs requires efficient deproteination methods to remove contaminating proteins.
    • Existing deproteination techniques may impact polysaccharide yield and integrity.

    Purpose of the Study:

    • To identify the optimal deproteination method for Dioscorea nipponica polysaccharides.
    • To evaluate and compare four different deproteination strategies.
    • To establish the best conditions for maximizing protein removal and polysaccharide recovery.

    Main Methods:

    • Four distinct deproteination methods were assessed.
    • The efficiency was determined by the ratio of protein removed to polysaccharides remaining.
    • Optimization involved varying papain concentration, pH, temperature, and incubation time, followed by the Sevage method.

    Main Results:

    • The papain-Sevage method demonstrated superior performance compared to other evaluated methods.
    • This method achieved a high deproteination rate of 91.24%.
    • Importantly, 80.12% of the Dioscorea nipponica polysaccharides were successfully retained.

    • Optimized conditions include 2.0% papain hydrolysis at pH 7.0 and 55°C for 122 minutes, followed by one application of the Sevage method.
    • This optimized protocol ensures efficient purification while preserving the integrity of the target polysaccharides.
