A novel orange-pigmented bacterium, Sphingomonas floccifaciens sp. nov., was discovered in Chinese permafrost sediments. This Gram-negative bacterium expands the known diversity within the Sphingomonas genus.
Area of Science:
- Microbiology
- Bacteriology
- Genomics
Background:
- Permafrost environments harbor unique microbial communities.
- The genus Sphingomonas is known for its metabolic versatility and presence in diverse environments.
Purpose of the Study:
- To isolate and characterize a novel bacterium from a subterranean sediment sample in China's Mohe permafrost area.
- To determine the phylogenetic and phenotypic characteristics of the novel isolate to establish its taxonomic position.
Main Methods:
- Isolation and cultivation of bacteria from permafrost sediment.
- Gram staining, motility, and sporulation tests.
- Optimal growth condition determination (temperature, pH, NaCl).
- Phylogenetic analysis using 16S rRNA gene sequences.
- DNA G+C content determination.
- Analysis of major ubiquinone, fatty acids, polar lipids, and polyamines.
Main Results:
- A Gram-negative, non-motile, non-sporulating, rod-shaped, orange-pigmented bacterium, strain FQM01T, was isolated.
- Strain FQM01T exhibited optimal growth at 25°C, pH 7.0, and 0% NaCl.
- Phylogenetic analysis placed strain FQM01T within the genus Sphingomonas, with Sphingomonas spermidinifaciens GDMCC 1.657T as the closest relative (97.6% similarity).
- The DNA G+C content was 66.9 mol%.
- Detailed chemotaxonomic analyses (ubiquinone, fatty acids, polar lipids, polyamines) supported its distinct identity.
Conclusions:
- Strain FQM01T represents a novel species within the genus Sphingomonas.
- The proposed name for this new species is Sphingomonas floccifaciens sp. nov.
- The type strain is FQM01T, with accession numbers CGMCC 1.15797T and KCTC 52630T.

Overview Of Cell Separation And Isolation
Cyclic Processes And Isolated Systems
In the case of a non-isolated system, the change in the internal energy is zero only if the process is cyclic. A thermodynamic process is considered cyclic if the system undergoes a series of changes and returns to its initial state.
Consider a cyclic process that returns to its initial state, undergoing a four-step process. The heat transfer along each...
