Purpose of the Study:

  • To develop novel EST-SSR markers for Pluchea indica.
  • To assess the genetic polymorphism within Pluchea indica populations.
  • To evaluate the cross-species utility of these markers in related Pluchea species.

Main Methods:

  • Transcriptome data from P. indica was used to design 40 primer pairs.
  • Seventeen primer pairs were successfully amplified and showed polymorphism.
  • Allele numbers and heterozygosity levels were calculated for each SSR locus.

Main Results:

  • 17 polymorphic EST-SSR markers were developed for P. indica.
  • Allelic diversity ranged from one to nine alleles per locus.
  • Observed and expected heterozygosity varied from 0 to 0.9 and 0 to 0.831, respectively.
  • Cross-amplification was successful in P. eupatorioides (16 loci), P. pteropoda (17 loci), and P. sagittalis (12 loci).

Conclusions:

  • The developed EST-SSR markers are effective for P. indica.
  • These markers can be applied to study the population genetics of P. indica.
  • The markers show potential for investigating genetic diversity in related Pluchea species.
