    Summary

    p-hydroxyamphetamine (p-OHA) induces head-twitches in mice, an experimental model for hallucination. This study suggests the central serotonergic system plays a key role in this p-OHA-induced effect.

    Background:

    • Head-twitches in mice serve as an experimental model for studying hallucinations.

    Purpose of the Study:

    • To investigate the involvement of the central serotonergic system in p-OHA-induced head-twitches.
    • To elucidate the mechanism underlying p-OHA-induced head-twitching behavior.

    Main Methods:

    • Administered p-hydroxyamphetamine (p-OHA) intracerebroventricularly (i.c.v.) to mice in a dose-dependent manner.
    • Examined the effects of simultaneous serotonin administration and pretreatment with serotonergic antagonists (p-chlorophenylalanine, cyproheptadine, dimethothiazine) on p-OHA-induced head-twitches.

    Main Results:

    • p-OHA administration resulted in dose-dependent head-twitches lasting 20-80 minutes, with peak effects at 30-40 minutes.
    • Co-administration of serotonin significantly increased the number of head-twitches.
    • Pretreatment with p-chlorophenylalanine, cyproheptadine, or dimethothiazine reduced the incidence of head-twitches.

    Conclusions:

    • The findings suggest that the central serotonergic system is involved in the mechanism of p-OHA-induced head-twitches.
    • The serotonergic system appears to exert an excitatory influence on this behavior.
