Simian virus 40 DNA replication: functional organization of regulatory elements.

G J Lee-Chen, M Woodworth-Gutai

    Summary

The structural organization of simian virus 40 (SV40) regulatory regions significantly impacts DNA replication efficiency. Specifically, G+C-rich 21-base-pair repeats enhance replication, but their effect is dependent on position, dose, and orientation.

    Area of Science:

    • Molecular Biology
    • Virology
    • Genetics

    Background:

    • Simian virus 40 (SV40) DNA replication is a complex process regulated by specific DNA sequences.
    • The regulatory region of SV40 contains G+C-rich 21-base-pair (bp) repeats that are known to influence replication efficiency.

    Purpose of the Study:

    • To investigate the impact of structural organization, including position, dose, and orientation, of the 21-bp repeats on SV40 DNA replication.
    • To examine the interplay between the 21-bp and 72-bp repeat regions in modulating SV40 replication.

    Main Methods:

    • Transfection experiments using SV40 DNA constructs with varying arrangements of the 21-bp repeat region relative to the origin of replication (ori).
    • Single and mixed transfection assays were employed to assess replication efficiency under different conditions.

    Related Experiment Videos

    Main Results:

    • The enhancing effect of the 21-bp repeats on SV40 DNA replication is dependent on their position, dose, and orientation.
    • Inverted orientation of the 21-bp repeats reduced enhancement by approximately 50%.
    • Relocating the 21-bp repeats upstream of the ori abolished enhancement, while placement downstream resulted in no replication.
    • The enhancing effect observed in single transfections was significantly amplified in mixed transfections.
    • The 72-bp repeat region enhanced SV40 replication in the presence of 21-bp repeats, but exhibited a cis-acting inhibitory effect in a competitive environment with 21-bp repeats.

    Conclusions:

    • The structural organization of the SV40 regulatory region, particularly the 21-bp repeats, is critical for efficient DNA replication.
    • Positional, dose, and orientation effects of the 21-bp repeats significantly modulate SV40 replication.
    • Interactions between different repeat elements (21-bp and 72-bp) play a complex role in regulating SV40 DNA replication, with context-dependent enhancement and inhibition observed.
