We developed a biosensor by attaching horseradish peroxidase (HRP) to reduced graphene oxide (RGO) for detecting hydrogen peroxide (H2O2) in serum. This novel RGO-HRP sensor shows promise for clinical diagnostics.
Area of Science:
- Biomaterials Science
- Nanotechnology
- Analytical Chemistry
Background:
- Hydrogen peroxide (H2O2) is a key biomarker in various physiological and pathological processes.
- Developing sensitive and selective biosensors for H2O2 detection in complex matrices like serum is crucial for diagnostics.
Purpose of the Study:
- To tether horseradish peroxidase (HRP) onto reduced graphene oxide (RGO) for effective H2O2 sensing in serum.
- To explore a green synthesis route for RGO and advanced functionalization techniques.
Main Methods:
- Reduced graphene oxide (RGO) synthesized via a green reduction of graphene oxide (GO) using carrot extract.
- Covalent functionalization of RGO using electrochemical amination with amine-terminated PAMAM dendrimers.
- Post-functionalization of HRP onto the RGO surface via glutaraldehyde linkage.
Main Results:
- Successful synthesis and characterization of functionalized RGO using spectroscopic, microscopic, and electrochemical methods.
- Demonstrated application of the HRP-tethered RGO biosensor for H2O2 detection in blood serum samples.
- Verified the stability and performance of the developed biosensor.
Conclusions:
- The HRP-tethered RGO platform offers a robust and sensitive approach for H2O2 sensing in serum.
- The employed green synthesis and functionalization strategy is adaptable for other carbonaceous materials.
- This work paves the way for advanced electrochemical biosensors in clinical applications.
