Certain brain-derived neurotrophic factor (BDNF) gene variations are linked to fibromyalgia (FM) risk and symptom severity. Specific BDNF polymorphisms and haplotypes may offer protection against FM development and influence its clinical presentation.
Area of Science:
- Neuroscience
- Genetics
- Rheumatology
Background:
- Brain-derived neurotrophic factor (BDNF) is implicated in fibromyalgia (FM) pathophysiology.
- Altered BDNF levels are observed in FM patients' blood and cerebrospinal fluid.
Purpose of the Study:
- To investigate the association between BDNF gene polymorphisms and FM susceptibility.
- To explore the relationship between BDNF gene polymorphisms and FM symptom severity.
Main Methods:
- Genotyping of 10 positions in the BDNF gene.
- Analysis of allele and genotype frequencies in 409 FM patients and 423 healthy controls.
- Haplotype analysis of BDNF gene variations.
Main Results:
- Significant differences in BDNF rs11030104 allele and genotype frequencies between FM patients and controls.
- The GG genotype and G allele of rs11030104 showed a protective effect against FM.
- The TT genotype of BDNF rs12273539 was associated with increased FM susceptibility.
- Certain BDNF haplotypes demonstrated a protective effect against FM.
- Specific BDNF genotypes and haplotypes correlated with FM symptom presentation.
Conclusions:
- This study is the first to link BDNF gene polymorphisms to FM.
- Specific BDNF single-nucleotide polymorphisms and haplotypes are associated with FM susceptibility.
- BDNF gene variations contribute to the development and symptoms of FM.
