Mechanical vibration (MV) enhanced the bone-forming effects of intermittent parathyroid hormone (iPTH) therapy in rats. However, MV did not maintain bone mass after iPTH treatment ended.
Area of Science:
- Bone biology and osteoporosis research
- Mechanobiology and therapeutic interventions
- Endocrinology and metabolic bone disease
Background:
- Intermittent parathyroid hormone (iPTH) is a bone-forming therapy for severe osteoporosis, limited to 24 months.
- Antiresorptive drugs are required post-iPTH to preserve newly formed bone tissue.
- Mechanical load, particularly vibration, shows potential for preventing bone loss.
Purpose of the Study:
- To assess if whole-body mechanical vibration (MV) can enhance iPTH's anabolic effects.
- To determine if MV can inhibit bone resorption after iPTH treatment cessation in estrogen-deficient rats.
Main Methods:
- Ovariectomized (OVX) and sham-operated rats were used, divided into seven treatment groups.
- Treatments included vehicle, MV alone, iPTH alone, iPTH combined with MV, and sequential iPTH and MV.
- Bone mineral density (BMD), body composition, histomorphometry, and biomechanical properties were analyzed.
Main Results:
- Mechanical vibration (MV) potentiated the anabolic effects of intermittent parathyroid hormone (iPTH) on cancellous bone.
- BMD increased in all iPTH-treated groups; PTH+MV showed the highest bone volume (BV/TV).
- Vibration applied after iPTH treatment improved tibial bone strength but did not maintain overall bone mass.
Conclusions:
- Mechanical vibration can amplify the bone-building effects of iPTH.
- Mechanical vibration alone is insufficient to maintain bone mass after iPTH treatment discontinuation in this model.
- Combined or sequential therapies warrant further investigation for osteoporosis management.
