A gene is a stretch of DNA that serves as the blueprint for functional RNAs and proteins. mRNA Stability and Gene Expression

The structure and stability of mRNA molecules regulates gene expression, as mRNAs are a key step in the pathway from gene to protein. In eukaryotes, the half-life of mRNA varies from a few minutes up to several days. mRNA stability is essential in growth and development. Background:

Invasive pathogens like Sphaerulina musiva pose significant threats to ecosystems and economies.
Traditional breeding programs for disease resistance are time-consuming, often taking decades.

Purpose of the Study:

To accelerate the identification of disease resistance genes in Populus trichocarpa.
To pinpoint genetic loci associated with resistance and susceptibility to Sphaerulina musiva.

Main Methods:

Genome-wide association mapping was performed on approximately 1,000 resequenced Populus trichocarpa trees.
Trees were individually challenged with the invasive fungal pathogen Sphaerulina musiva.
Candidate gene functions were supported by allele analysis, transcriptomics, binding assays, and overexpression studies.

Main Results:

Three loci associated with resistance to Sphaerulina musiva were identified.
These resistance loci are predicted to encode a membrane-bound L-type receptor-like kinase and two receptor-like proteins.
A susceptibility locus was identified, predicted to encode a G-type D-mannose-binding receptor-like kinase.

Conclusions:

The identified genes play a crucial role in Populus trichocarpa's response to Sphaerulina musiva infection.
This research demonstrates a faster approach to breeding for disease resistance in forest trees.
The findings provide valuable genetic resources for developing resistant Populus genotypes.
