Evidence that colicin X is microcin B17.

J L San Millán, R Kolter, F Moreno

    Journal of Bacteriology
    |June 1, 1987

    Summary

Antibiotic activities previously called colicin X are likely the same as microcin B17, a DNA replication inhibitor from Escherichia coli. This finding is supported by comparing production, action, and resistance patterns between the two.

    Area of Science:

    • Microbiology
    • Molecular Biology
    • Genetics

    Background:

    • Microcin B17 is a peptide antibiotic produced by Escherichia coli that inhibits DNA replication.
    • Colicin X has been described as a separate antibiotic activity.
    • The genetic basis for colicin X production is not fully understood.

    Purpose of the Study:

    • To investigate the relationship between microcin B17 and colicin X.
    • To determine if colicin X is identical to microcin B17.

    Main Methods:

    • Comparison of antibiotic production conditions.
    • Analysis of antibiotic mode of action.
    • Testing cross-immunity of producer strains and cross-resistance of resistant mutants.
    • DNA homology analysis of plasmids encoding colicin X and microcin B17.

    Main Results:

    • Antibiotic activities previously named colicin X are likely identical to microcin B17.
    • Identical production conditions, mode of action, cross-immunity, and cross-resistance were observed.
    • Plasmids encoding colicin X show significant DNA homology with the microcin B17-producing region of pMccB17.

    Conclusions:

    • Colicin X activity is identical to microcin B17.
    • The genes responsible for colicin X production are homologous to those of microcin B17.
    • This finding clarifies the identity of these antibiotic substances and their genetic basis.
