Inflammatory cytokines are elevated in dry eye disease (DED) patients, with specific differences found between Sjogren syndrome aqueous tear deficiency (SS-ATD), non-Sjogren syndrome aqueous tear deficiency (NSS-ATD), and meibomian gland dysfunction (MGD) subtypes. These findings suggest cytokines as diagnostic markers and therapeutic targets for DED.
Area of Science:
- Ophthalmology
- Immunology
- Biochemistry
Background:
- Dry eye disease (DED) is a multifactorial condition affecting millions globally.
- Understanding the inflammatory profiles of DED subclasses is crucial for targeted treatment.
- Tear inflammatory cytokines play a significant role in DED pathogenesis.
Purpose of the Study:
- To analyze tear inflammatory cytokines in different dry eye disease (DED) subclasses using Luminex technology.
- To compare cytokine levels between Sjogren syndrome aqueous tear deficiency (SS-ATD), non-Sjogren syndrome aqueous tear deficiency (NSS-ATD), and meibomian gland dysfunction (MGD) patients.
- To validate Luminex assay's utility for multiplex cytokine analysis in DED.
Main Methods:
- Collected tear samples from 45 DED patients (SS-ATD, NSS-ATD, MGD) and 15 normal controls.
- Measured concentrations of 11 inflammatory cytokines using Luminex assay.
- Validated Luminex results with ELISA assays.
Main Results:
- Most inflammatory cytokines were elevated in DED patients compared to controls.
- Significantly higher levels of TNF-α, IL-6, IL-8, and IL-12P70 were observed in DED patients.
- Distinct differences in IL-8 and TNF-α levels were found among SS-ATD, NSS-ATD, and MGD patient groups.
Conclusions:
- Elevated inflammatory cytokine levels are characteristic of DED.
- Specific cytokine profiles differentiate DED subclasses (SS-ATD, NSS-ATD, MGD).
- Inflammatory cytokines show potential as diagnostic biomarkers and therapeutic targets for DED.
