Summary

Adolescent females with anorexia nervosa (AN) show reduced brain gray matter and hippocampus volume. Most hippocampal subfields were smaller in AN patients, indicating significant structural changes beyond general brain volume loss.

Background:

  • Anorexia nervosa (AN) is associated with reduced cerebral gray matter (GM) volume.
  • Previous research indicates reduced hippocampus volume in AN patients.
  • Hippocampal subfields (HS) have distinct functions and may be differentially affected by neuropathology.

Purpose of the Study:

  • To investigate hippocampal subfield (HS) volumes in adolescent females with restrictive anorexia nervosa (AN) compared to healthy controls (HC).
  • To determine if volume reductions in HS are independent of global brain volume changes.
  • To explore relationships between HS volumes and clinical covariates like depression and anxiety.

Main Methods:

  • Utilized FreeSurfer v6.0 for brain volume extraction and HS segmentation.
  • Compared 30 adolescent females with AN to 28 age-matched healthy females (HC).
  • Analyzed group differences in GM, white matter (WM), whole hippocampus, and 12 HS volumes.

Main Results:

  • AN patients exhibited significantly lower total GM and total hippocampal volume compared to HC.
  • No significant group difference was observed in white matter (WM) volume.
  • Volume reduction was detected in 11 out of 12 HS, with most remaining significant after adjusting for global brain volume.

Conclusions:

  • Adolescent females with AN demonstrate reduced volumes in most hippocampal subfields.
  • This reduction in HS volume appears more extensive than the overall global cerebral volume loss observed in AN.
  • Clinical correlates, including depression and anxiety scores, showed significant relationships with whole hippocampus and several HS volumes in AN patients.
