We developed 18F-olaparib, a novel radiotracer for imaging Poly(ADP-ribose) polymerase (PARP) activity. This tracer shows promise for noninvasive tumor imaging and monitoring radiation damage in cancer patients.
Area of Science:
- Nuclear medicine
- Radiochemistry
- Cancer therapy
Background:
- Poly(ADP-ribose) polymerase (PARP) inhibitors are crucial in cancer treatment.
- Noninvasive imaging of PARP activity is needed for patient selection and therapy monitoring.
Purpose of the Study:
- To develop and evaluate 18F-olaparib, a novel PET imaging agent for PARP.
- To assess the potential of 18F-olaparib for monitoring radiation-induced DNA damage.
Main Methods:
- Copper-mediated 18F-radiofluorination of aryl boronic esters to synthesize 18F-olaparib.
- In vitro studies using PARP-1 expressing cells.
- PET imaging in mice bearing PARP-1 expressing tumors, with and without irradiation.
Main Results:
- 18F-olaparib selectively uptake in PARP-1 expressing cells.
- Tumor uptake of 18F-olaparib correlated with PARP-1 expression in vivo.
- Tumor uptake increased by 70% after irradiation, demonstrating sensitivity to DNA damage.
Conclusions:
- 18F-olaparib is a novel PET tracer for imaging PARP activity.
- 18F-olaparib enables noninvasive monitoring of radiation damage in tumors.

What is Gene Expression?
Gene expression is the process in which DNA directs the synthesis of functional products, that is, proteins. Cells can regulate gene expression at various stages. It allows organisms to generate different cell types and enables cells to adapt to internal and external factors.
Genetic Information Flows from DNA to RNA to Protein
A gene is a stretch of DNA that serves as the blueprint for functional RNAs and proteins. Since DNA is made up of nucleotides and proteins consist of amino...

Radiological Investigation III: Pulmonary Angiogram and PET Scan
Pulmonary Angiogram
A Pulmonary Angiogram is an invasive procedure involving injecting a contrast medium through a catheter threaded into the pulmonary artery or the right side of the heart to visualize the pulmonary vasculature. Computed Tomography (CT) scans have mainly replaced this...
Expressing Solution Concentration
Concentrations may be quantitatively assessed using a wide variety of measurement units, each convenient for particular applications. Molarity (M) is a useful concentration unit for many applications in chemistry.
