    Background:

    • Optimal sites for arterial or venous infusion and sampling in isotopic tracer studies remain undetermined.
    • Understanding plasma and tissue enrichment relationships is crucial for accurate metabolic flux analysis.

    Purpose of the Study:

    • To determine the relationship between plasma and tissue enrichment using arterial-venous (av) and venous-arterial (va) infusion/sampling modes.
    • To identify the optimal sampling strategy for isotopic tracer studies.

    Main Methods:

    • Adult dogs received constant infusions of [3-13C]lactate, [1-13C]leucine, and 14C-bicarbonate.
    • Simultaneous blood samples were collected from the vena cava and aortic arch; breath and tissue samples (skeletal muscle, liver, kidney, gut) were also obtained.

  • Isotopic enrichments were analyzed using liquid scintillation counting and gas chromatography-mass spectrometry.

    • The va mode resulted in plasma enrichment 30-40% higher than tissue enrichment for lactate and leucine.

    • Plasma alpha-[13C]ketoisocaproate (KIC) enrichment, a leucine metabolite, better reflected tissue leucine enrichment and was site-insensitive.

    • The arterial-venous (av) mode is optimal for isotopic tracer studies.
    • The venous-arterial (va) mode's limitations can be addressed by using metabolic products like KIC or employing circulatory models for calculation.
